VZV ORF49 (UniProtKB – Q77NP3) is a virion tegument protein. Sadaoka et al found that complex formation between ORF44 and ORF49 is required for the efficient production of infectious progeny virus. Only rabbit sera was available to analyse the ORF49 before we generated VZV mAbeom (Lenac Rovis et al, Virology 377:289 –295). Our monoclonal antibody VZ 49.07 confirmed in IF that ORF49 is localized to the Golgi network and other membranes in VZV infected cells. The ORF49 has calculated molecular weight of 8.9 kDa and our VZ 49.07 confirmed slightly bigger protein of 15 kDa in western blot. Its HSV-1 ortholog, UL11 is a myristylated virion protein. Our anti-VZV ORF49 antibody VZ 49.07 works in ELISA (on immunogen), IF (on VZV infected cells) and each LOT is validated for Western blot (of VZV infected cell lysates, described in J Virol. 2013 Jun;87(12):6943-54.).
