VZV ORF40, the major capsid protein, is a homomultimer that self-assembles to form an icosahedral capsid (UniProtKB – Q4JQT5). Its HSV-1 ortholog is UL19. Nagel et al (J. Virol. March 2011 vol. 85 no. 5 2276-2287) detected 10 VZV transcripts in latently infected human trigeminal ganglia in the following order of prevalence: ORF 63 (69%), ORF 29 (38%), ORF 4 (31%), ORF 62 (27%), ORF 11 (27%), ORF 68 (23%), ORF 40 (15%), ORF 43 (8%), ORF 41 (4%), and ORF 57 (4%). The ORF40 has calculated molecular weight of 115 kDa and our monoclonal antibody VZ 40.1.03 confirmed a 115 kDa protein in western blot. Our anti-VZV ORF40 antibody VZ 40.1.03 works in ELISA (on immunogen) and each LOT is validated for Western blot (of VZV infected cell lysates, described in J Virol. 2013 Jun;87(12):6943-54.).

VZV ORF21 encodes a 115 kDa protein that is essential for viral replication in cell culture and is also expressed during, but is not required for establishment of latency in dorsal root and cranial nerve ganglia.
VZV ORF27 encodes a nuclear egress lamina protein, that together with the nuclear egress membrane protein encoded by the product of VZV ORF27, most likely plays a role in the egress of assembled viral capsids from the nuclei of VZV infected cells to the cytoplasm.
